Is there a way to backup my settings/configuration?
I do NOT mean the books, I keep those on Dropbox
If I want to set my library up on a new computer, how can I back up my custom settings, custom search and replace regex's, plugins, etc. |

You might want to try calibre's builtin import/export tool under the library icon but that will also backup your library.
As for keeping your library in Dropbox? Do us all a favour and don't come back here and complain when your library gets corrupted. I've gotten tired of linking to: I am getting errors with my calibre library on a networked drive/NAS?. |

Copy the configuration folder (on a Windows computer, it's usually located in User/AppData/Roaming and is named calibre - copy the entire folder) and paste to the same location on your new computer.

Dropbox is file sharing between people and/or devices. Not a real backup solution.

In Calibre, "Preferences->Miscellaneous->Open configuration folder" will show you the folder with your config settings.
