How to make a paragraph stronger for screen readers?

Hello,
I have a book that has pull quotes. These are paragraphs that in print are bigger text, color and font than the regular text. These are short paragraphs that the author thinks the reader needs to keep in mind.

For a screen reader that is all just a simple paragraph, the same as all the others.

Can I use <em> or <strong> or some other coding so that the screen reader will emphasize that paragraph?

A screen reader worth it's electrons should emphasize text within <em>....</em> or <strong>....</strong> tags. A few screen readers will use different voices for <strong> and <em> but those are rare.
