Combining all your interests:
- literary
- upbeat (humorous)
- sci-fi
and you wind up with Jasper Fforde's "Thursday Next".

From the Amazon review:

I've only read the first book, and I'm not enough of a bibliophile to understand all the literary references it contains, but I loved this book, and plan to buy the rest, if I can ever find them on sale.
