As for your problem, I've got a couple of questions for you:

1. Did you actually download all Chinese dictionaries? (AFAIK, there's one for Simplified Chinese and one for Traditional Chinese.)

2. Does dictionary lookup work with other Traditional Chinese books that you didn't create yourself?

AFAIK, it's not possible to set sub-languages in Calibre. However, you can use Java Mobipocket Metadata editor to change the language code to a variety of Chinese that requires Traditional Chinese characters, for example, Taiwan:

Try changing the language code to see if it makes a difference.
